Description
FOXO4-DRI is a synthetic peptide designed to disrupt the interaction between the transcription factor FOXO4 and the tumor suppressor protein p53. It is primarily studied for its role in cellular senescence, a biological state in which damaged or aged cells permanently stop dividing but remain metabolically active. Accumulation of senescent cells is associated with aging, tissue dysfunction, and chronic inflammation.
Research interest in FOXO4-DRI centers on its ability to selectively target senescent cells by interfering with FOXO4-mediated nuclear retention of p53. By disrupting this interaction, FOXO4-DRI promotes apoptosis specifically in senescent cells while sparing healthy, proliferating cells. This targeted mechanism has positioned FOXO4-DRI as a key research tool in aging biology and senescence-related disease models.

FOXO4-DRI interferes with the FOXO4–p53 protein–protein interaction, a pathway that maintains senescent cells in a viable state by suppressing apoptosis.
